Keep in mind that you have to kill every boss, so you won't get healed in between floors. You don't really need to go for specific cards, since you only get 3-4 new cards anyway and they won't be enough to break the encounter on the first floor (apart from Soul Link and that soul link artifact), instead I would go for a few strategies that enable you to finish the run in its entirety. It is definitely a strategy to run regular Saffron runs until you get a broken deck and only then go for shopkeep, but I think this way here is faster and more fun. The other is that I think her health scaling is crazy and it is much harder to get her on higher floors unless you get lucky with a broken deck, which I don't like to rely on personally. This is for two reasons: first, she is most likely to break your run, so doing it on the first floor costs the least amount of time (about 5-6 minutes per run).

To complete the full genocide run, I start with default loadout and look for an early shopkeeper kill. If you don't know the patterns, chrono won't help too.
Also, if you are going for the full genocide run, I feel default is easier to complete the run at the end, especially if you kept your revive. If you know the patterns, the time stop is not that useful and as the others said, it is much easier to break the ice prison with default + you get one "freebie" if you mess up a timing severely, you can skip one entire attack with default. Ok, so in my experience, default loadout is easier than chrono. Aside from soul link cheese, which of course works but is kind of lame:
